Extracellular vesicle miRNA from cerebrospinal fluid as potential biomarkers for neurodegeneration

Abstract Background Extracellular vesicles (EVs) are a diverse group of small membranous particles classified by protein and nucleic acid content, size, method biogenesis, cellular origin. The need for predictive biomarkers neurodegenerative disease is critical early detection potential therapeutic intervention. We have previously identified multiple high ranking miRNAs in total cerebrospinal fluid (CSF) patients with Alzheimer's (AD). To investigate whether these released from cells within EVs, we developed pipeline to isolate analyze EV miRNA. Method EVs isolated size exclusion chromatography (SEC) CSF collected living donors. counts sizes determined nanoparticle tracking analysis (NTA). Immunoblots used characterize composition assess purity. Transmission electron microscopy (TEM) structural characteristics. RNA using commercial based methods. Quantitative PCR (qPCR) performed TaqMan Low Density Array (TLDA) miRNA cards. Result isolation protocol here capable isolating pure the appropriate composition. Following qPCR, more than half AD were observed population. Conclusion This suitable assessing content lumbar CSF. A large number appear correlate well published data CSF, indicating role cargo as diseases such AD.
